I smoked a waterfall before a trip once and I was tripping serious balls very quickly. Be careful of overdoing it on your first time, especially if you get bored in the come-up which can be lengthy. Having something fun to do really cuts the anxiety rather than sitting around waiting for something to happen.

Tolerance builds quick, man. Taking 2 a few days after taking 1 will just lead to a mediocre experience. Don't chase an ideal trip. Wait a while, plan something cool to do and enjoy. I have taken similar doses of the same acid in different situations and each time it feels different. Highly recommend doing something outside in the day with a friend you feel comfortable with. As for effects of regular use, I went through a period of tripping semi-regularly and I can't say I noticed anything particularly. Maybe a slight bit of dissociation, but nothing terrible.
